AOD-9604 is a synthetic fragment corresponding to amino acids 177–191 of human growth hormone — specifically, the region responsible for hGH's fat-metabolising (lipolytic) activity, isolated from the parts of the hormone that drive tissue growth and raise blood glucose.
A key 2001 study published in Endocrinology found that both full-length hGH and its AOD9604 fragment could induce weight loss and increase lipolytic sensitivity in obese mice following chronic treatment, and — critically — that this lipolytic action was not mediated through the same growth-promoting pathway as the intact hormone. Because AOD-9604 doesn't meaningfully engage the IGF-1 axis the way full hGH does, it avoids the glucose and insulin-sensitivity effects associated with growth hormone therapy.
AOD-9604 went on to complete six randomised, placebo-controlled human trials involving over 900 participants assessing safety and tolerability, though development as a licensed weight-loss drug was discontinued in 2007 after its largest Phase IIb trial failed to show statistically significant weight loss over placebo — a reminder that a sound mechanism doesn't always translate into a clinically decisive result.
